On the side of a barn which is covered with latex? What should I use if I start with bare wood?

Sherwin Williams A-100 OIL primer. Then for your mural use either a-100 or superpaint. All are Sherwin Williams products.

Oil would probably be your bet bet because it takes the most time to dry. Plus, you are going to be outside so the paint would dry even faster.
